Detection Chemistry
The Discovery XT system can be used as a stand-alone ISH staining system, as an IHC stand-alone staining unit, or as a combination ISH/IHC unit. The IHC software allows the researcher to choose among different pre-packaged detection chemistries:
Alkaline phosphatase detection using NBT/BCIP chemistry (blue)
Alkaline phosphatase detection using a Fast Red/Napthol chemistry( red)
Peroxidase detection using DAB chemistry (brown)

The Discovery XT system can be used for in situ
hybridization using DNA, RNA, and oligo probes performed on tissue sections,
cell preparations, or
chromosome spreads, using either chromogenic or fluorescent detection methods.

The in situ hybridization application software has features that can accommodate the programming of the various procedural steps required for in situ hybridization such as pretreatments, probe application, denaturation, hybridization, and stringency washes and detection.

Because of the unique technical principles of the Ventana staining system, hybridization can be performed with all probe types available to the research field. Ventana's Discovery XT system is well suited for the study of gene expression profiles or detection of viruses.
in situ Hybridization Steps:
The probe application can be performed either automatically through a dispenser or manually by the researcher. The denaturation and hybridization temperatures can be set from 37 ฐC to 95ฐC and from 37ฐC to 70ฐC respectively, while hybridization time may be extended up to 12 hours. The instrument allows the user to choose up to three stringency washes, with selectable incubation times and temperatures.
Microarrays*
Ventana offers optimized reagents and protocols for standardized hybridization of a labeled target to DNA microarrays* printed on glass slides.
The current manual methods for microarray* hybridization are inconsistent, labor intensive and time consuming. Typically, the results are less than optimal with variable signal intensities, non-uniformity due to diffusion limitation, high probe concentration requirements, and low throughput due to extended hybridization times. Ventana's Discovery XT system and the ChipMapฎ Kit optimized reagents promote uniform spreading and maximize the efficiency of hybridization conditions to provide excellent sensitivity. The proprietary formulations minimize non-specific binding, providing quality results with high signal-to-noise ratio and exceptional specificity. Uniform hybridization on the entire microarray* surface yields greater consistency, reduces slide-to-slide variation, and improves reproducibility and reliability.
